Sexual Assault Evidence

For a variety of reasons, some victims of sexual assault provide vaginal samples more than 24-36 hours after the incident.  The ability to obtain an autosomal STR profile of the semen donor from the living victim diminishes rapidly as the post coital interval is extended.  We have developed various enhancement strategies to extend the post coital interval (up to 9 days) from which a genetic profile of the semen donor can be obtained.  We attribute our success to a number of factors that significantly improve the sensitivity and specificity of the analysis, including the development of a Y chromosome targeted pre-amplification strategy.  The results of this work are likely to have a significant impact on the investigation of rape cases and the time frame in which sexual assault evidence is collected.

U.S. Y-STR Database

The significance of a Y-STR match in casework is estimated by reference to a reliable and relevant database of Y-STR profiles. NCFS currently maintains the U.S. Y-STR Database (funded by the National Institute of Justice, NIJ), a population database intended for use in estimating Y-STR haplotype population frequencies for forensic casework purposes. Currently, an 11 to 23-locus profile can be searched, which will soon be expanded to 27 loci to include all loci used in commercially available Y-STR kits. As of the last update (August 2013), the database contained 29, 754 haplotypes. The database is continually expanded with haplotype data contributions from operational, academic and commercial laboratories throughout the U.S.

Body Fluid Identification

DNA typing to identify the donor of transferred biological traces deposited at the crime scene or on a person or implement is now a routine practice worldwide. This represents potentially critical ‘source level’ information for investigations. However, a DNA profile from the perpetrator doesn’t reveal the circumstances by which it got transferred. This contextual information or ‘activity level’ is important because the deposition of the perpetrator’s biological material requires some behavioral activity on behalf of the individual that results in its transfer from the body. The consequences of different modes of transfer of the DNA profile may dramatically affect the investigation and prosecution of the crime. Conventional methods of body fluid identification use labor-intensive, technologically diverse techniques that are costly in terms of time and sample.  For some frequently encountered body fluids (e.g. saliva, vaginal secretions, menstrual blood and skin) no definitive identification technique exists. Therefore, there is a critical need for the development of novel strategies for the identification of forensically relevant biological fluids.  A pattern of gene expression is produced in body fluid secretions and tissues that uniquely defines each cell type present. This is evidenced by the presence and relative abundance of specific RNA species. We have developed a number of highly specific RNA profiling assays, using both messenger RNA (mRNA) and microRNA (miRNA) biomarkers, for body fluid and tissue identification that employ a variety of commonly used analytical platforms.

Trace Biological Evidence

In forensic casework analysis, it is sometimes necessary to obtain genetic profiles from increasingly less amounts of biological material left behind by persons involved in criminal offenses. Forensic casework evidentiary items sometimes contain less than 100 pg of template DNA, which is equivalent to approximately 15 diploid or 30 haploid cells. The presence of such low template DNA (LTDNA) samples could be due to several factors including damaged or degraded DNA, oligospermic or aspermic individuals, extended interval post coital samples, or other trace biological evidence such as ‘touch DNA’. Since only a small amount of genetic material is present in these samples, recovery of genetic profiles is challenging using standard DNA analysis methods. Therefore, novel strategies for the improved analysis of trace biological evidence need to be developed. We are investigating the use of different strategies (e.g. laser capture micro-dissection, micro-manipulation, micro-volume PCR) to improve profile recovery from trace biological evidence.

The donor of a body fluid recovered from the crime scene is identified by comparing the crime scene DNA profile to profiles obtained from reference samples obtained from known individuals.  However, in the absence of appropriate comparison reference samples, the DNA profile per se presently provides no meaningful information to investigators, with the notable exception of gender determination. A number of other physically recognizable characteristics of an individual may be amenable to molecular genetic analysis and these include skin-, hair- and eye- color, stature (height and weight), age and facial morphology. Our focus is on the development of novel strategies for the determination of ethno-geographic ancestry and the biological age of an individual.

Enhanced DNA Analysis

A goal of our research efforts is to aid in the development of novel DNA technologies to make DNA analysis quicker, better or cheaper. Our main focus areas include mixture de-convolution and the development and validation of “rapid” DNA screening and analysis techniques and instrumentation (R-DNA).

Mixtures are frequently encountered in forensic DNA analysis due to the involvement of multiple persons during the commission of certain crimes.  Mixtures are also often found associated with ‘touch DNA’ or with other crime scene samples due to unavoidable contamination with previously deposited DNA. Analysis and interpretation of these admixed DNA samples is often quite complex and therefore extremely challenging even more so if the number of donors exceeds two. A limited number of tools are available to operational forensic laboratories in order to aid in the interpretation or handling of mixtures. Our work has focused on the development of novel strategies for the de-convolution of body fluid mixtures containing non-distinguishable cell types (e.g. epithelial cells and leukocytes) including the use of binomial sampling strategies combined with quantitative computer- based interpretation systems.

The next revolution in forensic DNA analysis, apart from the application of next generation sequencing technology, is the deployment of R-DNA systems.  The concept behind this is that actionable forensic intelligence can be obtained in situ (at the crime scene, booking station etc) by instrumentation that permits ‘sample-in results out’ analysis within an hour or so.  We are assisting in the testing and evaluation of some of these systems, in particular the RapidHIT 200 (IntegenX) and paraDNA (LGC) platforms.  The RapidHIT system is capable of providing a full CODIS autosomal STR profile whereas the paraDNA system is principally designed as a cost-effective screen or intelligence system.

Fundamental Biochemical Processes

One of the limiting factors with DNA typing technology is that sometimes DNA isolated from physiological stains recovered from the crime scene is so damaged as to be intractable to analysis.  However, current knowledge of the biochemical nature and extent of the DNA damage in biological stains of forensic importance is rudimentary. We have investigated the basic chemistry of the dried state, specifically that of the most common expected type of chemical reactions, namely hydrolysis and oxidation to determine the rates of such reactions under different environmental conditions. We have also characterized the molecular lesions that prevent the ability to obtain a standard DNA profile in damaged DNA recovered from crime scene physiological stains with a view to determining possible DNA repair strategies.  Subtle physicochemical changes to the structure of the hemoglobin molecule in the dried state in bloodstains have been used by us to develop a simple VIS spectroscopic method for the estimation of the age of the bloodstain (i.e. time since deposition (TSD)).

As technology becomes increasingly integral to our daily lives, it's no surprise that crime-solving has advanced to near-futuristic levels. From retinal scanning to trace evidence chemistry, modern forensic technologies are so sophisticated that they seem straight out of a science-fiction thriller.

Due to these advancements, the field of forensic science is rapidly growing in the U.S. One clear indicator of this growth is the rising demand for forensic science technicians. According to the Bureau of Labor Statistics (BLS), there will be a 13% increase in jobs for forensic science technicians nationally from 2022 to 2032. The BLS attributes this growth to the development of new forensic science techniques, which have enhanced the availability and reliability of objective forensic information. Consequently, courts and law enforcement agencies are hiring additional staff to utilise these techniques and analyse data for use in trials.

Top 10 Modern Forensic Science Technologies 2024

Since 2020, there have been over 22,000 scientific publications detailing new advances in forensic science. These include groundbreaking technologies as well as novel methods for analyzing evidence within established areas. These innovations are making it increasingly challenging for criminals to avoid detection by forensic scientists. Here is the list:

NIST aims to cultivate trust in the design, development, use and governance of Artificial Intelligence (AI) technologies and systems in ways that enhance safety and security and improve quality of life. With AI and Machine Learning (ML) changing how society addresses challenges and opportunities, the trustworthiness of AI technologies is critical. Trustworthy AI systems are those demonstrated to be valid and reliable; safe, secure and resilient; accountable and transparent; explainable and interpretable; privacy-enhanced; and fair with harmful bias managed. The agency’s AI goals and activities are driven by its statutory mandates, Presidential Executive Orders and policies, and the needs expressed by U.S. industry, the global research community, other federal agencies,and civil society.

NIST’s AI goals include:

  • Conduct fundamental research to advance trustworthy AI technologies.
  • Apply AI research and innovation across the NIST Laboratory Programs.

High groundwater depletion risk in South Korea in 2080s

Groundwater is literally the water found beneath the Earth's surface. It forms when precipitation such as rain and snow seeps into the soil, replenishing rivers and lakes. This resource supplies our drinking water. However, a recent study has alarmed the scientific community by predicting that approximately three million people in currently untapped areas of Korea could face groundwater depletion by 2080.

A research team, led by Professor Jonghun Kam from Division of Environmental Science and Engineering and Dr. Chang-Kyun Park from the Institute of Environmental and Energy Technology (currently working for LG Energy Solution) at Pohang University of Science and Technology (POSTECH), used an advanced statistical method, to analyze surface and deep groundwater level data from 2009 to 2020, revealing critical spatiotemporal patterns in groundwater levels. Their findings were published in the international journal "Science of the Total Environment."

Groundwater is crucial for ecosystems and socioeconomic development, particularly in mountainous regions where water systems are limited. However, recent social and economic activities along with urban development have led to significant groundwater overuse. Additionally, rising land temperatures are altering regional water flows and supplies, necessitating water policies that consider both natural and human impacts to effectively address climate change.

In a recent study, researchers used an advanced statistical method called "cyclostationary empirical orthogonal function analysis (CSEOF)" to analyze water level data from nearly 200 surface and deep groundwater stations in the southern Korean Peninsula from 2009 to 2020. This analysis helped them identify important spatiotemporal patterns in groundwater levels.

The first and second principal components revealed that water level patterns mirrored recurring seasonal changes and droughts. While shallow-level groundwater is more sensitive to the seasonality of precipitation than the drought occurrence, deep-level groundwater is more sensitive to the drought occurrence than seasonality of precipitation. This indicates that both shallow-level and deep-level groundwater are crucial for meeting community water needs and mitigating drought effects.

The third principal component highlighted a decline in groundwater levels in the western Korean Peninsula since 2009. The researchers projected that if this decline in deep groundwater continues, at least three million people in untapped or newly developed areas, primarily in the southwestern part of the peninsula, could face unprecedented groundwater level as a new normal (defined as groundwater depletion) by 2080. If the research team's predictions are correct, the impact would be particularly severe in drought-prone, untapped areas where groundwater is heavily relied upon.

Professor Jonghun Kam of POSTECH stated, "By leveraging long-term, multi-layer groundwater level data on Korea and advanced statistical techniques, we successfully analyzed the changing patterns of deep- and shallow-level groundwater levels and predicted the risk of groundwater depletion." He added, "An integrated national development plan is essential, one that considers not only regional development plans but also balanced water resource management plans."

NSF invests $36M in computing projects that promise to maximize performance, reduce energy demands

The U.S. National Science Foundation is awarding $36 million to three projects selected for their potential to revolutionize computing and make significant impacts in reducing the carbon footprint of the lifecycle of computers. Funding for the projects comes from the NSF Expeditions in Computing (Expeditions) program, an ambitious initiative that supports transformative research poised to yield lasting impacts on society, the economy and technological advancement. Projects funded by Expeditions are characterized by their ambition and potential for transformation, leveraging advances in computing and cyberinfrastructure to accelerate discovery and innovation across various domains of science and engineering. 

"We are thrilled to announce these visionary projects that will advance environmental responsibility and foster innovation in the field of computing," said Dilma DaSilva, acting assistant director for the NSF Directorate for Computer and Information Science and Engineering (CISE). "Congratulations to these pioneering teams whose research will forge new pathways in computational decarbonization and in revolutionizing operating system design with machine learning.    

The 2024 NSF Expeditions awardees   

NSF Expeditions in Computing: Carbon Connect--An Ecosystem for Sustainable Computing . Led by Harvard University, this multi-institutional, five-year research initiative will lay the foundations for sustainable computing, with a focus on reducing the environmental impact of computer systems. This shift toward sustainability could spark a transformation in how computer systems are manufactured, allocated and consumed, leading to a more responsible and sustainable approach to advancing computing technologies. By redefining the way computer scientists consider environmental sustainability, Carbon Connect will establish new standards for carbon accounting in the computing industry, thereby influencing future energy policy and legislation. 

Collaborators on this project include the University of Pennsylvania, the California Institute of Technology, Carnegie Mellon University, Cornell University, Yale University and The Ohio State University. 

NSF Expeditions in Computing for Computational Decarbonization of Societal Infrastructures at Mesoscales . Led by the University of Massachusetts Amherst, this project will develop the new field of computational decarbonization, (CoDec), which focuses on optimizing and reducing the lifecycle of carbon emissions of complex computing and societal infrastructure systems. CoDec will tackle interdependencies across multiple aspects of infrastructure, including computing, transportation, buildings and the electric power grid. Through innovative sensing approaches, optimization methods grounded in theory and artificial intelligence, and software-defined interfaces, CoDec seeks to automate and coordinate carbon-efficiency optimizations across time, space and sectors. These efforts will enable scientific discoveries in decarbonization while supporting sustainable growth, advancing technology and strengthening national security.   

Collaborators of this project include Carnegie Mellon University, the Massachusetts Institute of Technology, the University of Chicago, UCLA and the University of Wisconsin-Madison. 

NSF Expeditions in Computing: Learning Directed Operating System--A Clean-Slate Paradigm for Operating Systems Design and Implementation . Led by The University of Texas at Austin, this project aims to revolutionize the design of operating systems (OSes) by integrating advanced machine learning (ML) into resource management. Current OSes employ rigid, manually designed approaches for allocating hardware resources among running applications. This inflexibility makes it hard to adapt to evolving application needs and hardware, leading to inefficiency and poor performance. This research will develop a learning-directed operating system with intrinsic intelligence and auto-adaptation, enabling ML-driven resource management that optimizes performance and efficiency and requires minimal human intervention. By fundamentally rethinking OS design with ML at its core, this research has the potential to significantly improve the energy efficiency of cloud computing, enable real-time edge computing applications and create innovative computing devices.

Established in 2008, the NSF Expeditions awards represent some of the largest investments provided by the CISE directorate. Pioneering work funded by the program includes the Robobee Project and CompSustNet .  

Expeditions projects focus on creating transformative technologies, methodologies and infrastructure that can be adopted by the broader research community, industry or society at large. The program emphasizes the translation of research outcomes into practical applications, thus driving advancements in computer science and its real-world applications.
